Measurement of absorption spectrum

Summary Absorbance measurement is used to determine the concentration of light-absorbing solutions and gases in optical transmission media. The unit signal of absorbance is proportional to the molar absorption, optical path and sample concentration.

USB4000-UV-VIS spectrometer is suitable for 200-850nm absorbance measurement. The spectrometer is preset with a # 1 grating with a peak efficiency of 300nm. This configuration provides suitable resolution (~ 1.5nm FWHM) for the absorbance measurement of most solutions. The built-in OFLV-200-850 anti-diffraction filter can shield the second- and third-order diffraction effects. If it is not shielded, it will produce an unclean peak in the absorption spectrum. It is recommended to use the DH2000-BAL deuterium-tungsten halogen combination lamp as the light source. Although the DH2000 light source is cheaper than the light source, the D-alpha line of the UV light source in the DH2000 is not used in some specific experiments because of the lack of filter technology May cause misunderstanding.


Sampling optics For absolute absorbance measurement, a 1-cm optical path CUV-UV tube holder and CV-Q-10 quartz test tube can be used. For relative absorbance measurement, a directly attached USB accessory, immersion probe and flow cell can be used. We recommend using QP400-025-SR advanced anti-exposure fiber as the illumination and reading fiber. Adopt the STAN-ABS absorbance measurement standard that complies with NIST standards to ensure accurate and reliable results.
