Recently, Cell Research published the latest research results of Lilin Research Group of Institute of Biochemistry and Cell on Dishevelled protein involved in the regulation of NF-kB signaling pathway. Dishevelled protein is a very key regulatory protein in the Wnt signaling pathway. Not only does it play a role in the transmission of Wnt signal from the cell membrane to the intracellular, but it has recently been found to participate in the formation of the transcription complex of the classic Wnt signal in the nucleus. The research was supported by projects from the Ministry of Science and Technology, the National Fund Committee and the Shanghai Science and Technology Commission. In this study, Ding Ning and Ye Yanger of Li Lin's research group discovered that Dishevelled protein interacts with NF-kB core molecule p65 in the nucleus in a way that does not depend on Wnt signaling, thereby negatively regulating p65-mediated NF-kB activity. Further research shows that Dishevelled inhibits the binding of p65 to the promoter regions of certain target genes (including some anti-apoptotic genes), thereby inhibiting the anti-apoptotic effect of NF-kB.
