Previous literature has shown that there is a relatively large D / H fractionation between fat and water in most organisms. This fractionation comes down to the effect of isotopes on fat biosynthesis. In this paper, we report the D / H fractionation between the fat and water of four bacteria (phylum Proteobacteria), and the results show that the fluctuation in a single organism can reach 500 ‰. This change cannot be attributed to fat biosynthesis, because there is no obvious change in these pathways, nor can it be attributed to the D / H ratio of the culture medium. More importantly, the D / H of fat / water changes systematically with metabolism: chemical autotrophic growth (almost reaching -200 to -400 ‰), photosynthetic autotrophic growth (-150 to -250 ‰), non- Autotrophic organisms, organisms that use sugar as a medium (0 to -150 ‰), and non-autotrophic organisms, use the TCA cycle (-50 to -200 ‰). We speculate that the D / H ratio of fat is largely controlled by biosynthetic NADPH, not the fat biosynthesis pathway itself. Our results indicate that different metabolic pathways produce NADPH—and indirectly affect the isotopic composition of fat. If so, the δD value of fat may become an important biochemical cycle tool connecting fat and energy metabolism, and may provide more supplementary information through 13C in the carbon fixation pathway.
